Gene Expression in Escherichia coli

Essentially all genes can be expressed by using recombinant methods. Gene expression in Escherichia coli is the most efficient system.

Keywords: gene expression; Escherichia coli; promoter; recombinant protein

Figure 1. Schematic illustration of a vector and a DNA construct, and a typical process of gene expression in Escherichia coli. RBS, ribosomal-binding site; MCS, multiple cloning site.
Figure 2. Expression of a truncated mouse mannose-binding protein (MBP) in Escherichia coli. E. coli cells containing the plasmid for MBP expression were cultivated at 37°C with shaking. After cells were grown to the desired density, isopropylthio--d-galactoside (IPTG) was added to induce expression. Aliquots were removed before induction (0 h), 1, 2, 3, 4, 9, 17 h after induction, as indicated. The cells were lysed and the cell lysates were resolved using sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS-PAGE), and stained with Coomassie Blue. S, protein mass standards.
